A iniciativa recebeu men\u00e7\u00e3o honrosa no 7th Brazilian Association for Bioinformatics and Computational Biology, em 2011, e o pr\u00eamio de melhor apresenta\u00e7\u00e3o no 2\u00ba Workshop do Programa Interunidades de P\u00f3s-Gradua\u00e7\u00e3o em Bioinform\u00e1tica da USP, em 2012.<\/p>\n<div title=\"Texto\">\n<p style=\"text-align: justify;\">O protocolo re\u00fane m\u00e9todos experimentais e computacionais para obten\u00e7\u00e3o de modelos tridimensionais de prote\u00ednas multidom\u00ednio, mol\u00e9culas que representam dois ter\u00e7os de todo proteoma dos seres eucari\u00f3ticos e que est\u00e3o relacionadas a diversos eventos org\u00e2nicos, como processos de sinaliza\u00e7\u00e3o celular, prolifera\u00e7\u00e3o do c\u00e2ncer, diabetes, insufici\u00eancia card\u00edaca, entre muitas outras. O projeto faz parte do doutorado do t\u00e9cnico em desenvolvimento do LNBio Rodrigo Honorato, aluno do programa de p\u00f3s gradua\u00e7\u00e3o em bioinform\u00e1tica da USP.<\/p>\n<p style=\"text-align: justify;\">O conhecimento da estrutura dessas prote\u00ednas \u00e9 fundamental para desvendar suas fun\u00e7\u00f5es, processos bioqu\u00edmicos por elas mediados e intera\u00e7\u00f5es com outras mol\u00e9culas. Os m\u00e9todos comumente utilizados para resolver estruturas moleculares muitas vezes n\u00e3o s\u00e3o adequados para as prote\u00ednas multidom\u00ednios, j\u00e1 que elas n\u00e3o formam cristais est\u00e1veis para serem submetidos \u00e0 difra\u00e7\u00e3o de raios X, e s\u00e3o grandes demais para serem estudadas por resson\u00e2ncia magn\u00e9tica nuclear. O protocolo MAD foi criado exatamente para superar essa dificuldade, com a vantagem de poder ser adaptado ao estudo de intera\u00e7\u00f5es entre diferentes prote\u00ednas.<\/p>\n<p style=\"text-align: justify;\">O pesquisador do LNBio\u00a0<a href=\"https:\/\/lnbio.cnpem.staging.wpengine.com\/oliveira\/\" target=\"_self\" data-cke-saved-href=\"https:\/\/lnbio.cnpem.staging.wpengine.com\/oliveira\/\" rel=\"noopener\">Paulo S\u00e9rgio Lopes de Oliveira<\/a>, um dos respons\u00e1veis pelo desenvolvimento do MAD, avalia que as informa\u00e7\u00f5es fornecidas poder\u00e3o levar a medicamentos mais eficazes. \u201cO conhecimento das estruturas das prote\u00ednas multidom\u00ednios permite desenvolver f\u00e1rmacos com alta especificidade, diferentes dos atuais, que muitas vezes agem sobre toda uma fam\u00edlia de prote\u00ednas e causam diversos efeitos colaterais\u201d, explica.<\/p>\n<p style=\"text-align: justify;\"><strong>Estruturas<\/strong><\/p>\n<p style=\"text-align: justify;\">Os pesquisadores trabalham agora para acurar as estruturas obtidas por meio desse protocolo. Para isso, prote\u00ednas multidom\u00ednios j\u00e1 conhecidas, como a super\u00f3xido desmutase e a neurotoxina botul\u00ednica, est\u00e3o sendo submetidas ao MAD, e os resultados obtidos, comparados \u00e0s estruturas j\u00e1 validadas. Os comparativos apontam para o sucesso do m\u00e9todo.<\/p>\n<p style=\"text-align: justify;\">Pesquisadores do LNBio \u2013 laborat\u00f3rio que integra o\u00a0<a href=\"https:\/\/www.cnpem.staging.wpengine.com\/\" target=\"_blank\" data-cke-saved-href=\"https:\/\/www.cnpem.staging.wpengine.com\/\" rel=\"noopener\">Centro Nacional de Pesquisa em Energia e Materiais (CNPEM)<\/a>, organiza\u00e7\u00e3o social supervisionada pelo Minist\u00e9rio da Ci\u00eancia, Tecnologia e Inova\u00e7\u00e3o (MCTI) \u2013 j\u00e1 fizeram uso do protocolo com \u00eaxito. Pesquisa publicada na Nature Chemical Biology, publicada em janeiro de 2012, valeu-se tamb\u00e9m desse m\u00e9todo para caracterizar o dom\u00ednio FERM da enzima card\u00edaca FAK, quinase de ades\u00e3o focal. Os resultados desse trabalho apresentaram uma oportunidade de modula\u00e7\u00e3o dessa enzima, por meio desse dom\u00ednio, que poder\u00e1 auxiliar no combate \u00e0 hipertrofia card\u00edaca.<\/p>\n<p style=\"text-align: justify;\">Outra pesquisa do LNBio, publicada no The Journal of Biological Chemistry, em dezembro, empregou o MAD para estudar as intera\u00e7\u00f5es entre a prote\u00edna relacionada ao c\u00e2ncer oral, ADAM17, e a tiorredoxina, uma pequena prote\u00edna com fun\u00e7\u00e3o antioxidante. Os resultados mostraram que a atividade da ADAM17 \u00e9 inibida pela liga\u00e7\u00e3o com essa pequena mol\u00e9cula, o que sugere um alvo terap\u00eautico para o c\u00e2ncer oral.
